Circulating chemerin levels are determined through circulating platelet counts in nondiabetic Taiwanese people: A bidirectional Mendelian randomization study

چکیده
Background and aimsPlatelet count (PLT) is a predictor of metabolic inflammation-related disorders. Platelets can release prochemerin, which acts as link between coagulation inflammation innate adaptive immunity. The causal effect PLT circulating chemerin level has not been elucidated.MethodsNondiabetic participants with samples in the Taiwan Biobank were recruited for genome-wide association study (GWAS) based on (17,037 participants) levels (3887 participants). A bidirectional Mendelian randomization (MR) was conducted to determine levels.ResultsFor GWAS PLT, 11 gene loci found have significance. For levels, two loci, RARRES2 HLADQA2-HLADQB1, Age, sex, body mass index, leukocyte count, hemoglobin, mean blood pressure, hemoglobin A1C, serum total bilirubin, aspartate aminotransferase, triglyceride, low-density-lipoprotein cholesterol estimated glomerular filtration rate, be independently associated through stepwise regression analysis. MR revealed weighted genetic risk scores (WGRSs) significantly by using two-stage least-square method multivariate analysis (p = 0.0031), no significant WGRSs noted. Sensitivity further violation exclusion-restriction assumption PLT-determining genotypes levels.ConclusionsThrough analysis, our data that determined PLT. Circulating intermediates future

A Mendelian Randomization Study of Circulating Uric Acid and Type 2 Diabetes.
We aimed to investigate the causal effect of circulating uric acid concentrations on type 2 diabetes risk. A Mendelian randomization study was performed using a genetic score with 24 uric acid-associated loci. We used data of the European Prospective Investigation into Cancer and Nutrition (EPIC)-InterAct case-cohort study, comprising 24,265 individuals of European ancestry from eight European ...
متن کاملCirculating levels of chemerin and adiponectin are higher in ulcerative colitis and chemerin is elevated in Crohn's disease.
BACKGROUND Chemerin is an adipokine that stimulates chemotaxis of cells of the innate immune system. Inflammatory bowel disease (IBD) is linked to an impaired immune response and, therefore, we hypothesized that systemic chemerin may be altered in IBD patients. METHODS Serum was collected from patients with Crohn's disease (CD, 230 patients), ulcerative colitis (UC, 80 patients), and healthy ...
متن کاملCausal Assessment of Serum Urate Levels in Cardiometabolic Diseases Through a Mendelian Randomization Study.
BACKGROUND Although epidemiological studies have reported positive associations between circulating urate levels and cardiometabolic diseases, causality remains uncertain. OBJECTIVES Through a Mendelian randomization approach, we assessed whether serum urate levels are causally relevant in type 2 diabetes mellitus (T2DM), coronary heart disease (CHD), ischemic stroke, and heart failure (HF). ...
متن کاملCirculating interleukin-6 and cancer: A meta-analysis using Mendelian randomization
Interleukin-6 (IL-6) plays a contributory role in the progression and severity of many forms of cancer; it however remains unclear whether the relevance between circulating IL-6 and cancer is causal. We therefore meta-analyzed published articles in this regard using IL-6 gene -174G/C variant as an instrument. Seventy-eight and six articles were eligible for the association of -174G/C variant wi...
متن کاملCausal relevance of circulating high-density lipoprotein cholesterol with cancer: a Mendelian randomization meta-analysis
We summarized published data on the associations of apolipoprotein E (APOE) gene ε2/ε3/ε4 polymorphism with both cancer risk and circulating lipid profiles, aiming to examine the causal relevance between lipids and cancer risk. Article identification and data abstraction were conducted in duplicate and independently by two authors. Data were analyzed by STATA software. Twenty-five articles that...
